چکیده

AimsTo evaluate the effects of fetal glucose infusion in late gestation on mRNA expression and protein abundance molecules involved regulation cardiac growth metabolism.Main methodsEither saline or was infused into sheep from 130 to 140 days (d) (term, 150 d). At d gestation, left ventricle tissue samples were collected. Quantitative real-time RT-PCR Western blot used determine key signalling within heart.Key findingsAlthough intra-fetal increased plasma insulin concentrations, there no change pathways that regulate proliferation, hypertrophy, apoptosis fibrosis heart. Cardiac Solute carrier family 2 member 1 (SLC2A1) decreased by infusion. Glucose both Peroxisome proliferator activated receptor alpha (PPARA) peroxisome gamma (PPARG). However, PPAR cofactors with response elements. Furthermore, did not impact 5 oxidative phosphorylation complexes electron transport chain.SignificanceDespite a 10-day doubling present study suggests ventricle, growth, development metabolism are relatively unaffected.

Fetal morphine metabolism and clearance are constant during late gestation.

Fetal metabolism significantly contributes to the clearance of drugs from the fetus. To understand how the changes in fetal metabolism expected in late gestation alter fetal drug clearance, serial measurements of morphine metabolism were made in the fetal baboon over the latter third of gestation. Clearance and metabolism were evaluated in the context of fetal growth, onset of labor, and the ad...

Exogenous GH infusion to late-gestational fetal sheep does not alter fetal growth and metabolism.

The role of GH in the regulation of fetal growth and metabolism in late gestation is not well defined. The aim of this study was to determine the effects of exogenous GH infusion on fetal growth and feto-placental metabolism in the normally growing late-gestation fetal sheep. Eleven fetuses received pulsatile GH infusion (3.5 mg/day) for 10 days while 12 control fetuses received vehicle. The GH...

Effects of intrafetal IGF-I on growth of cardiac myocytes in late-gestation fetal sheep.

Intrafetal insulin-like growth factor (IGF)-I promotes cardiac hypertrophy in the late-gestation fetal sheep; whether these effects are sustained is unknown. IGF-I was infused for 4 days at 80 microg/h from 121 to 125 days of gestation, and its effects at 128 days, 3 days after the infusion stopped, were determined by comparison with untreated fetal sheep. After IGF-I treatment, fetal weights w...

Plasma leptin concentration in fetal sheep during late gestation: ontogeny and effect of glucocorticoids.

The ontogeny and developmental control of plasma leptin concentration in the fetus are poorly understood. The present study investigated plasma leptin concentration in chronically catheterized sheep fetuses near term, and in neonatal and adult sheep. The effect of glucocorticoids on plasma leptin in utero was examined by fetal adrenalectomy and exogenous cortisol or dexamethasone infusion. In i...
